چکیده
Colorectal cancer (CRC) is the third most common in world known for its poor recurrence-free prognosis. Previous studies have shown that it closely linked with stem cells (CSCs), which self-renewal potential and capacity to differentiate into diverse populations. Nanog an important transcription factor functions maintain proliferation of embryonic cells; however, many recent also highly expressed cells. To investigate whether plays a crucial role maintaining stemness colorectal CSCs, RNA interference was used downregulate expression CRC cell line, EpCAM+CD44+HCT-116 (CCSCs). We examined anti-tumor function vitro vivo, using small interfering RNA. Our results revealed mRNA level CCSCs higher than HCT-116 found depletion inhibited promoted apoptosis CCSCs. In addition, invasive ability markedly restricted when silenced by Furthermore, we silencing decreased tumor size weight improved survival rate tumor-bearing mice. conclusion, these findings collectively demonstrate Nanog, cells, key development growth, may serve as marker prognosis novel effective therapeutic target treatment CRC.

RNAi-mediated RPL34 knockdown suppresses the growth of human gastric cancer cells
An increasing body of evidence suggests that ribosomal proteins may have ribosome-independent functions and may be involved in various physiological and pathological processes. To examine the role of ribosomal protein L34 (RPL34) in cancer transformation, we assessed its expression in gastric cancer cell lines and found it highly expressed. We further used lentivirus-mediated small interfering ...
متن کاملEffects of CD133 Silencing on Survival and Migration of HT-29 Colorectal Cancer Cells
Background: Colorectal cancer (CRC) is attributed as one of the most common malignancies worldwide. CD133 molecule, as a pentaspan transmembrane glycoprotein, confers stem cell-related characteristics, including self-renewal and multi-directional differentiation capability. CD133 plays important roles in the progression of CRC by conferring apoptotic resistance and migration ab...
متن کاملEffects of RNAi-mediated Smad4 silencing on growth and apoptosis of human rhabdomyosarcoma cells.
Transforming growth factor-beta (TGF-beta) signals through membrane-bound heteromeric serine/threonine kinase receptors. Upon ligand binding, TGF-beta activates intracellular Smad proteins and regulates proliferation and apoptosis in various cell types. To demonstrate the effects of TGF-beta/Smad signal on growth and apoptosis of human embryonal rhabdomyosarcoma (RMS) cells, a strategy of RNAi-...
متن کاملSilencing of RhoA and RhoC expression by RNA interference suppresses human colorectal carcinoma growth in vivo
BACKGROUND RhoA and RhoC have been proved to be over-expressed in many solid cancers, including colorectal cancer. The reduction of RhoA and RhoC expression by RNA interference (RNAi) resulted growth inhibition of cancer cells. The present study was to evaluate the effect of silencing of RhoA and RhoC expression by RNAi on growth of human colorectal carcinoma (CRC) in tumor-bearing nude mice in...
متن کاملInhibition of TMEM16A Expression Suppresses Growth and Invasion in Human Colorectal Cancer Cells
Metastasis leads to poor prognosis in colorectal cancer patients, and there is a growing need for new therapeutic targets. TMEM16A (ANO1, DOG1 or TAOS2) has recently been identified as a calcium-activated chloride channel (CaCC) and is reported to be overexpressed in several malignancies; however, its expression and function in colorectal cancer (CRC) remains unclear. In this study, we found ex...
